As SpliceBio is growing, we're looking for exceptional professionals to be part of our team, supporting the development of gene therapies for diseases for which currently no treatment exists. We are now hiring a Scientist Vector engineering for AAV Gene Therapy. In this capacity you will be responsible for leading an ambitious scientific vision and strategy for internal AAV gene therapy vector genome research. Moreover the successful candidate will be empowered to develop and implement the strategic view for the next generation of gene therapy platforms and programs, working in close collaboration with the Head of Translational and Chief Development Officer.
The successful candidate will have a strong background and hands-on experience working in molecular biology, AAV vector engineering, genetic engineering techniques e.g. CRISPR, PCR, cloning, FACS, transfection, transduction, and assay development and also have strong knowledge in the regulatory DNA elements that can be modified to improve transgene expression. Production and purification of AAVs is a plus.
He/She will work collaboratively with cross-functional teams and external third partners, ensuring the highest standards of data quality and regulatory compliance are met throughout the development process.
Reporting to our Head of Translational, this is what you'll be working on:
• Lead AAV construct design-related activities, drive innovation in AAV cargo design.
• Ensure the successful design of the AAV skeleton to improve expression of the transgene in the desired target cells.
• Hands-on work in the cloning and screening of the AAV vectors (e.g. cell culture transfection and transduction and characterization of the constructs).
• Perform and/or oversee sample analysis and data interpretation.
• Works closely with external partners on AAV viral vector optimization, ensuring the timely completion of projects and the delivery of high-quality data.
• Author and review technical and scientific documents including but not limited to study protocols and reports, validation documents, SOPs, and test methods.
• Actively participates as a member of multifunctional project teams. Collaborate with cross- functional teams, including research and development, as well as external third parties, to ensure seamless integration of data into product development.
Requirements For a successful fulfilment of this position, we are interested in candidates meeting the following requirements:
• PhD in virology, virus bioengineering, gene/cell therapy, molecular biology or related field.
• At least 4 years of practical experience in the industry/academic field, with a focus on AAV gene therapy.
• Experience with AAV plasmid design and cloning and Mammalian cell/tissue culture (including transfection and/or transduction).
• Experience with a variety of cellular and molecular biology techniques such as qPCR/ddPCR, flow cytometry, Western Blot, etc.
• Strong organizational skills, able to deal with a large volume of samples.
• Strong analytical and problem-solving skills, with the ability to troubleshoot complex technical issues and develop innovative solutions.
• Proficiency in data analysis, interpretation and reporting using relevant software tools.
• Confident and clear presentation of data, verbally and through written reports.
• Motivated to learn and develop new methods and techniques.
• Expertise in managing relationships with external partners, including CROs, and overseeing outsourced activities will be a plus.
• Production and/or experimental use of viral vectors (especially AAV) will be a plus.
• Excellent communication and interpersonal skills, with the ability to work effectively in a team-oriented, fast-paced environment.
• Fluency in English, written and spoken.

SpliceBio's platform technology, developed over 20 years in the Muir Lab at Princeton University, allows for efficient delivery of large genes with adeno-associated vectors (AAV). Genes are split into fragments to be then put back together to the full-length protein to be delivered.
In early 2022, the company completed the largest ever series A financing in the Spanish biotech sector; an oversubscribed €50 million finance round co-led by UCB Ventures and existing shareholder Ysios Capital and the participation of new investors New Enterprise Associates, Gilde Healthcare, Novartis Venture Fund and existing backer Asabys Partners which seeded the company in 2020 with Ysios. This financing round validates SpliceBio's approach of exploiting protein splicing to develop next generation gene therapies. The company is addressing one of the key limitations of AAV, being their limited cargo capacity.
